How does glutathione mediate fast, coordinated G1s?
The mechanism through which nuclear GSH regulates the G1 to S transition is unknown. Several possibilities exist including regulating nuclear reactive oxygen species, altering chromatin as a post-translational modification of histones, or by altering the species of nuclear proteins by targeting some for degradation.
The Lee lab is using synthetic approaches to manipulate nuclear GSH content and pairing that with proteomic, metabolomic, and next generation sequencing approaches to comprehensively profile the regulatory landscape of high- and low-GSH nuclei in order to determine a link between GSH levels and cell cycle regulation.
